The apolipoprotein E (APOE) gene is the major genetic source of the common form of Alzheimer disease developed later in life. The APOE-4 variation increases the risk for developing Alzheimer’s and decreases the average age when symptoms may first appear. APOE-4 accounts for only one part of the genetic risk, however, a family history of dementia, regardless of APOE-4 status, can also increase the risk for developing the disease. The true mechanism by which APOE affects Alzheimer risk has not been determined, although many scientists have explored the relationships between elevated cholesterol and blood sugar levels to Alzheimer.
